Sažetak
The activity of sperm superoxide dismutase (Cu, Zn SOD) is not fully eluciated in asthenozoospermia, characterised by normal spermatozoa concentration but with reduced motility. The aim of this study was evaluatethe SOD-like activity as well as in vitro effect of zinc on its activity in astheno and normozoospermia. SOD-like activity was determined in two Percoll fractions of 18 normozoospermic (>20x106 sperms/ml ; motility >40%)and 12 asthenozoospermic subjects (motility <40%).In vitro effect of zinc was assessed on unfractionated spermatozoa of 27 normo-and 18 asthenozoospermic males. SOD-like activity was measured using the RanSod test. SOD-like activity was significantly elevated in 40% Percoll fraction of asthenozoospermic when compared to normozoospermic samples (p<0.03). A nonsignificant increase was found in spermatozoa from 80% Percoll fraction (p<0.5). The incubation of sperm suspension (370C, 30 min, n=14)with increasing Zn concentration (0.1-3 mM/l)resulted in concentration-dependent decrease in SOD-like activity. The inhibitory effect of zinc (3 mM/l)was less pronounced in asthenozoospermic (p<0.001 ; n=18)than normozoospermic samples (p<0.0001 ; n=27). Results from this study indicate that asthenozoosperia is associated with both an increased sperm SOD-like activity and its stronger resistance to inhibitory effect of zinc, which might be relevant to the sperm function.
