Sažetak
Asthma has been known for its associations with different alleles within the 6p21 chromosomal region. This study aimed at characterizing the distribution of HLA class I and class II as well as TNFa and STR_MICA polymorphisms in specific asthma and atopy traits in the Croatian children population. One hundred and three patients and 150 controls were studied. Patients were analyzed by phenotypic axpression of symptoms (asthma with or without allergic rhinitis), total serum IgE, sensitization to allergens (Der p, Amb e and Dact g) and disease severity. Using appropriate statistical tests no significant associations were found for HLA-A, -C and –DRB1 loci after Bonfferoni correction has been applied. Positive association was demonstrated for: TNFa2 and asthma with associated rhinitis (pc=0.0247) ; HLA-B8, -DQB1*0603 and TNFa2 with more severe asthma (pc=0.046, 0.0238 and 0.0406 respecrtively) and STR_MICA-A5 with the groups of patients having simulatneously combined traits of greater severity: sensitivity to Amb e and/or Dact g in addition to Der p and more severe asthma (pc=0.0015), allergic rhinitis (pc=0.0005) or higher total IgE level (pc=0.0005) in adition. DPB1*0401 could confer resistance to asthma as a whole, to high serum IgE and to atopy (pc=0.0348, 0.0105 and 0.0294 respectively). Our data suggest that different loci within 6p21 as DQB1, TNFa and STR_MCA may predispose to the susceptibility to different clinical forms of asthma and atopy, especially to more severe ones.
