Sažetak
Serotonin (5-hydroxytriptamine) as a secondary plant product may have different functions in plant tissue. The accumulation of serotonin in seeds of walnuts is connected with the loss of moisture and vacuoles which are replaced by protein bodies in the cells during the ripening period. To prevent the rising of ammonia from permissible to toxic levels, the seeds of walnuts reduce their ammonia content by synthesizing serotonin. Methanolic extracts of black walnut"s and butternut"s cotyledons in four maturity stages was examined. Serotonin from extracts was separated on ion exchanger Amberlite CG-50 and detected by chromatographic and spectrofluorometric methods. Increasing level of serotonin during ripening was established in both species of walnuts. Serotonin was also in situ localised in cotyledons of walnuts using two histochemical regents: p-dimethylaminobenzaldehide in hydrochloric acid and ninhydrin in acetic acid.
