Sažetak
Serotonin (5-hydroxytryptamine, 5HT) is a biogenic amine involved in modulation of emotions and behaviors, as well as in the regulation of cardiovascular, respiratory, gastrointestinal and immunological functions. In order to investigate serotonin role in various physiological processes and pathological states, we developed an animal model termed Wistar Zagreb-5HT rat, which has constitutively altered both peripheral and central 5HT homeostasis. Model consists of two sublines developed by selective breeding of Wistar rats and characterized by increased (high- 5HT subline) or decreased (low-5HT subline) values of platelet 5HT transporter activity, platelet 5HT level, as well as plasma free 5HT. 5HT regulation of immune functions may be mediated either indirectly through central neuroendocrine pathways or directly through several distinct receptors expressed on immune cells. Our initial work on Wistar Zagreb-5HT rats demonstrated that animals from low-5HT subline have increased natural cytotoxic activity, as measured by natural killer (NK) activity and antibody-dependent cellular cytotoxicity (ADCC), in comparison to animals from high-5HT subline. On the other hand, immunization of the rats with sheep red blood cells (SRBC) resulted in ~ 50 % lower antibody titer in sera of low-5HT animals as compared to high-5HT animals. These results point to differences in adaptive immune response between sublines of rats with constitutively altered 5HT homeostasis and indicate opposite effects of 5HT on distinct immune reactions. Taking into account that symptomatic asthmatic patients have increased 5HT plasma levels that directly correlate to impaired pulmonary function, we suggest that Wistar Zagreb-5HT rat can serve as a useful model for investigation of pathophysiological role of 5HT in asthma and other allergic diseases.
