Sažetak
Peptidoglycan monomer (PGM), originally prepared by biosynthesis from culture fluids of penicillin-treated Brevibacterium divaricatum has been repeatedly shown to posses marked immunomodulating properties. Previously we reported that even better immunostimulating capacity has new metal analogue-PGM-Zn, whose immunocorrective activity in several states of immunodeficiencies (aging, halothane and jaundice induced immunosuppression) correlated with its ability to induce marked activation of hepatic and peritoneal macrophages. To elucidate the cytotoxic activities of macrophages, activated in vivo by PGM-Zn, in this study we assessed the effects of peritoneal exudate cells (PEC), obtained from mice pretreated with PGM-Zn (10 mg/kg i.p.) or with saline solution on concanavalin A (ConA) induced blastogenesis of autologous and syngeneic splenocytes, as well as, the cytotoxic effects of hepatic and splenic mononuclear lymphatic cells on the growth of Yac-1, the NK- sensitive T-cell line. Responsiveness of splenocytes to the mitogens and killing of Yac-1 cells were evaluated by a liquid scintillation counter and by FACS analysis, respectively. The data have shown that pretreatment of mice with PGM-Zn enhances the hepatic class II expression and phagocytic activities of PEC. These, PGM-Zn activated PEC were, however, unable to stimulate the Con-A blastogenesis of syngeneic splenocytes in contrast to those obtained from saline treated mice, suggesting that PGM-Zn had activated the macrophages with suppressive activity. Pretreatment with PGM-Zn increased also the cytotoxicity of hepatic and especially splenic cells NK, enhancing on dose depend manner the killing of Yac-1 cells. The data emphasize that PGM-Zn, as a new molecule, which contains two constituents with known immunomodulating activities, has interesting, new, antiproliferative and cytotoxic properties.
