Sažetak
Wild cherry (Prunus avium L.) is commercially interesting tree species in Yugoslavia but with limited supplies. Micropropagation of wild cherry was investigated. The dissected uninjured buds provided by different kinds of material (one- or five-year-old trees, suckers initiated on roots, and mature trees) were used as explants. The successful establishment of aseptic cultures was carried out by 1% chlorine preparation Izosan G and 1% mercuric chloride. A very good multiple shoot induction was obtained on Woody Plant Medium (Lloyd and McCown, 1980) without chlorine ions and with 60 mgl-1 NaFeEDTA supplemented with (µM): 2.2 BA, 2.5 IBA, 0.3 GA3, 2% sucrose and 0.9% Bacto-agar. The shoot multiplication rate was 4-7 for 4 or 5 weeks, and depended on the donor age. Elongated shoots were rooted in a high percentage after a pulse treatment with IBA solution (246 µM, 4 h) and planted in a sterile soil mixture. Plantlings were successfully adapted to greenhouse conditions.
