Sažetak
This study was undertaken to determine the extent to which organ of origin influence the PTEN protein immunohistochemical expression signature of endometrioid subtype of endometrial and ovarian tumours. Tissue samples of endometrioid subtype of 17 endometrial and 17 ovarian tumors were obtained from Department of Pathology, University Hospital of Split, Croatia. Immunohistochemical staining was performed on formalin fixed and paraffin embedded tissue samples using monoclonal PTEN antibody (R&D Systems, 1:40). Expression of PTEN protein was evaluated semiquantitatively by light microscopy (Olympus BX40). Immunohistochemical analysis revealed that PTEN protein expression was located only in the cytoplasm of tumor cells. Heterogeneous PTEN protein expression was noted in 24 % (4/17) of endometrioid endometrial tumor samples meaning that some cells within the gland or some glands within the tumor were negative for PTEN staining. Endometrioid ovarian tumors revealed heterogeneous PTEN protein expression in 12 % (2/17) of samples. Our results suggest that although endometrioid subtype of ovarian tumors bear close histological resemblance to endometrioid tumors of the endometrium they may arise from different genetic events determined by additional "organ – specific" factors.
