Sažetak
Ability of an organism to enhance or reduce some of the vital functions in response to various external and internal demands, and thus adapt to these new conditions, Sterling and Eyer (1988) defined as allostasis. However, frequent changes of physiological processes in terms of increased or decreased activity gradually exhaust the organs and tissues of the body and bring it into the state susceptibile to disease, so-called allostatic load (AL). Stress response is an integral part of allostasis since the same physiological and behavioural processes allow the survival of individuals in condition of sudden danger and its main mediators are the same that participate in the maintenance of the daily cycle of activity and rest (eg. adrenal hormones, neurotransmitters, cytokines). Four basic mechanisms by which stress can lead to allostatic load have been described: a) repeated challenges, b) failure to adapt to these challenges, c) failure to shut off the response after the challenge is over, and d) failure to establish an adequate response. According to this concept new summary measure of allostatic load (AL score) was proposed as an estimate of total burden to multiple systems “accumulated“ throughout life. This cumulative measure of allostatic load is based on determining the physiological values of ten (or more) biological measures (biomarkers) selected as primary and secondary mediators of allostasis. Such aggregate value has predictive significance for prevalence of cardiovascular disease and mortality. Based on our results and experience, we will suggest additional biomarker (NK activity) that may contribute to determination of allostatic load of immune system dysregulation.
