Sažetak
It has been proved that hyperglycaemia in diabetes mellitus have a toxic effect on a cell. The aim of this study was to investigate effect of different concentration glucose on a leakage LDH from Hep G2 cells. The Hep G2 cell line was obtained from American Type Culture Collection (ATCC). Hep G2 cells were grown in complete MEM with 2 mM glutamine, supplemented with 10 % fetal bovine serum, at 37 C, under a humidified atmosphere of 5 % CO2. The cells were incubated 4 hours with different concentration of glucose (20, 25, 30, 35, 40 mM) dissolved in complete medium. Leakage of LDH was measured as an index of cell-membrane damage. At the end of the experiment, the media and cells were collected and frozen on a - 80 C until analysis. The cells were harvested by scraping and suspended in 1.0 mL PBS/TWIN, than sonicated with an ultrasonicator for 15 sek. on ice. The LDH activities in the medium and in the cells were measured with method based on the recommendation of the IFCC on the Olympus AU 600 Analyser. Glucose in a concentration of 30 mM significantly enhanced the LDH leakage from Hep G2 cells, probably over glucose autooxidation. (LDH leakage in control Hep G2 cells = 25.6 % vs. LDH leakage in Hep G2 cells treated with 30 mM glucose = 31.3 % ; p = 0.036)
