Sažetak
PCR assays have been developed principally to assist investigations into the epidemiology of Giardia duodenalis, the only species in the Giardia genus that infects humans and other mammals. However, a reliable identification of all species is of practical importance, particularly when environmental samples are analyzed. In this work, we demonstrate the usefulness of the region spanning the 5.8S and the two flanking internal transcribed spacers (ITS) of the ribosomal operon, to genotype Giardia. Primers were designed to match conserved regions in the 3' end of the small subunit and in the 5' end of the large subunit of the ribosomal operon of G. duodenalis, G. ardeae and G. muris, and used to amplify this region from a large number (n= 50) of isolates. Sequence comparison and phylogenetic analysis showed that each Giardia species and each of the seven G. duodenalis assemblages have a distinct sequence. Isolates of Giardia microti collected from wild rodent species, and isolates of G. duodenalis assemblage B from several hosts showed the highest level of polymorphism in this region. Since the major subgroups within assemblage A and assemblage B can be identified, this assay can be used for molecular epidemiology studies.
