Sažetak
Statins are among the most frequently prescribed drugs for treating people with risk of cardiovascular diseases. They lower cholesterol levels by inhibiting HMG-CoA reductase, a rate-limiting step in cholesterol biosynthesis. Lovastatin is a natural product from Aspergilllus terreus, simvastatin and pravastatin are semi-synthetic, whereas fluvastatin, atorvastatin and rosuvastatin are completely synthetic compounds. Statins are formulated in the pharmacological active β -hydroxy acid form, while simvastatin and lovastatin are administered as lactone prodrugs and thereafter hydrolyze to active metabolites. Official methods for determination of atorvastatin, fluvastatin and rosuvastatin have not yet been approved in European Pharmacopoeia. Hence, simple and efficient methods for the determination of statins are highly required. The development of a rapid analytical procedure that will not be limited to the analysis of only one statin could be very useful. The separation of six statins was carried out in a fused-silica capillary having 50 μ m internal diameter and 32 cm total length (effective length to detector 23.5 cm). Detection was performed with a diode array detector at 237 nm. Base hydrolysis was used to open lactone rings of lovastatin and simvastatin in order to transform these compounds to the corresponding negatively charged hydroxyl acid forms enabling them to be determined in shorter analysis time. In method development the influence of pH was first studied using different buffers in the pH range 3.9-9.5. The borat buffer at pH 9.5 gave the best preliminary results since at this high pH the apparent mobilities of all six statin drugs were the highest. The addition of an organic modifier (10% v/v methanol) in the presence of SDS (25 mM) in the electrolyte solution played a key role in the separation of statin molecules. Good separation (Rs > 3.85) was achieved in a short analysis time allowing determination of six drugs within 5 min. The effect of capillary temperature and applied voltage were also examined. Optimised conditions were 30 º ; C and 23 kV. The newly developed method was applied for assays of all six statin drugs in commercial pharmaceutical product.
