Kao kontrola koriateni su akoljkaai izlagani u jezeru Bundek, kao i oni s referentne postaje Draganii. Kao pokazatelj oateenja DNA uslijed one iaenja koriaten je komet-test na hemocitima izlo~enih akoljkaaa. Odreeni su osnovni fizikalno-kemijski, mikrobioloaki i bioloaki pokazatelji kakvoe vode, a za neke postaje napravljena je i kemijska analiza sedimenta. Nakon 2 tjedna izlaganja rezultati komet- testa pokazali su statisti ki zna ajno vee oateenje DNA na veini potoka i na svim postajama na rijeci Savi u usporedbi s kontrolnim postajama. Sli an gradijent oateenja DNA izmeu postaja primijeen je i nakon 4 tjedna izlaganja, s tim da su u ovom slu aju razine oateenja bile neato viae. Naai rezultati opravdavaju kavezno izlaganje slikarske lisanke kao prikladan model za praenje utjecaja one iaenja okoliaa te potvruju primjenu komet testa kao pouzdanog pokazatelja u procjeni genotoksi nosti. Mussels were collected at the reference site Draganii and caged for a period of 2 and 4 weeks in autumn 2007, at twelve sites in the freshwater ecosystems of the Zagreb County. Seven sites were streams of different pollution intensity in Zagreb and further four polluted sites were in the river Sava. Mussels caged at the Bundek Lake, as well as the ones from the reference site Draganii were used as controls. Basic physical, chemical, microbiological and biological parameters of water quality were determined, and for some locations chemical analysis of water sediments was performed. Results of the comet assay after 2 weeks exposure indicated statistically significant increase in DNA damage in majority of the streams and all locations on the river Sava when compared to the control sites. Similar gradient of DNA damage between the investigated sites was observed after 4 weeks of exposure, although with somewhat higher DNA damage values. Our results indicated the caging exposure of painter s mussel as a suitable method for monitoring the impact of environmental pollution and we confirmed the usefulness of comet assay as a reliable biomarker in genotoxicity assessment.
