U ovom radu istra~en je genotoksi ni potencijal rijeke Kupe na prirodne populacije klena (Leuciscus cephalus L.) upotrebom komet- i mikronukleus-testa. Tijekom srpnja 2007. sakupljeni su uzorci riba s etiri postaje razli itog stupnja one iaenja du~ rijeke Kupe (Ozalj-Kamanje, Brod na Kupi, Karlovac, Sisak). Za sve su postaje odreeni fizikalno-kemijski parametri kakvoe vode, te napravljene kemijske analize metala, PKB-ova i PAU-ova u vodi, sedimentu i miaiu klena. Rezultati komet-testa pokazali su da je koli ina izmjerenog oateenja DNA eritrocita klena razmjerna one iaenju postaja utvrenim kemijskim analizama. Postaja Ozalj-Kamanje pokazala se kao ona s najmanjim genotoksi nim utjecajem, slijedi Brod na Kupi, dok su na one iaenim postajama Sisak i Karlovac izmjerena najvea oateenja DNA. Rezultati mikronukleus-testa nisu pokazali genotoksi ni u inak na one iaenim postajama. Ovi rezultati ukazuju na opravdanost primjene klena kao prikladne vrste u istra~ivanjima utjecaja one iaenja slatkovodnih ekosustava te potvruju primjenu komet- testa kao dobrog pokazatelja u procjeni genotoksi nosti. In this study genotoxic potential of the pollution of the river Kupa was examined in native populations of chub (Leuciscus cephalus L.) by the means of comet assay and micronucleus test. Fish were collected in July 2007 at four locations of different pollution intensity along the river Kupa (Ozalj-Kamanje, Brod na Kupi, Karlovac, Sisak). Chemical analysis of metals, PCB and PAH content in water sediments and chub as well as basic physical and chemical parameters of water quality have been determined at all locations. Results of the comet assay showed good correlation between the quantity of the DNA damage and the pollution status of the examined locations evaluated by the chemical analysis. The lowest level of DNA damage was detected at Ozalj-Kamanje site followed by the site Brod na Kupi. The highest levels of DNA damage were observed at the most polluted sites Sisak and Karlovac. Micronucleus test did not reveal any genotoxic effect at polluted sites. Our results indicated the applicability of feral chub as an appropriate species in aquatic genotoxicity testing and confirmed the usefulness of comet assay as a suitable tool for detection of pollution-related genotoxicity.
