Sažetak
Urban air pollutants, particularly benzene, toluene, etilbenzene, xilene (BTEX) and diesel exhaust particles contribute to the increasing incidence of many allergic and environmental diseases, as well as multiple chemical sensitivity or other related syndromes, what may result with an abnormal immune response based on environmental damage of lymphocyte subsets. The immune system has a capacity to differentiate innocuous and harmful foreign antigens, which are controlled by mechanisms of central and peripheral tolerance. The aim of this study was to examine alteration of cell mediate immunity and the role of cytolytic molecule perforin in different lymphocyte subpopulation in peripheral blood of inhabitants situated near gasoline industry compared with healthy donors. Venous blood samples were taken and phenotype of lymphocytes and perforin expression were analyzed using intracellular and surface immunofluoresecncy and flow cytometry (FACSCalibur). Statistical analysis was done in computer program Statistica 7.1 The results demonstrated that 17% of PBL in all examine group are perforin positive (total perforin is significantly lower in exposure than in control group). Percentages of double positive cells showed significant decrease in all examine lymphocyte subpopulations (CD3+ P+, CD4+P+, CD8+P+, CD16+P+ and CD56+P+ cells). Our data suggest that human perforin is differentially expressed in distinct sub-populations of peripheral blood lymphocytes of people exposure to gasoline and diesel, implicating the important role of perforin positive cells in innate immune response and cytotoxic activity of peripheral blood lymphocytes.
