Chemoprevention of tumor with natural components, including propolis and propolis related polyphenolic/flavonoid compounds has recently drawn attention as a strong antitumor approach to be used. In case of recurrent non-muscle-involved urothelial carcinomas after transurethral resection intravesical instillation of chchemotherapy or BCG immunotherapy have mostly been employed. In the current study we investigated the direct cytotoxicity of propolis preparations and propolis polyphenolic/flavonoid compounds on papillary urothelial carcinoma cells in primary culture assay. A dose-depedent cytotoxicity was found when cells were exposed to either ethanolic (EEP) or water soluble preparation (WSDP) of propolis ; the polyphenolic/flavonoid compounds such as caffeic acid (CA), naringin (Nar), chrysin (Chrys), and quercetin (QU) exerted cytotoxicity to cancer cells in various degrees. The most active test components in this respect were EEP and QU respectively when the dose of 300 µ ; g/ml of culture was used regardless to cancer grade. At any time of expositure (24, 48, 72 hours) the efficacy of EEP and QU was statisticaly significant (p<0.05 ; p< 0.01). On the other hand, any of test components used exibited no cytotoxicity on normal epithelial bladder cells in primary culture. The cytotoxicity of test components was caracterized by morphological changes specific for apoptosis. Data suggest that propolis and propolis related polyphenolic/flavonoid compounds may be a promising treatment for papillary urothelial bladder cancer used as an adjuvant after transurethral resection.
