We have shown previously ( M. Osmak et al., Anti-Cancer Drugs, 10:853-859, 1999), that from 10 new synthesized diazenecarboxamides (diazenes), diazene JK-279 was mostly effective: it reduced the cell survival of all 10 cell lines examined. The aim of the present study was to elucidate the molecular mechanisms involved in this phenomenon. Human cervical carcinoma HeLa cells were used as the experimental system. As diazenes may reduced the intracellular level of glutathione (GSH), the concentration of GSH was determined spectrophotometrically by the procedure developed by Tietze. The kinetic of apoptosis induction was followed after the treatment with JK-279 by fluorescent microscope for 72 h posttreatment period. The occurrence of apoptosis was confirmed by analyzing DNA fragmentation in agarose gel. Results show that diazene JK-279 reduced significantly the intracellular level of GSH ( up to 17 % of control level). It induced apoptosis, that could be detected even one hour after the treatment, as shown by DNA ladders. These results suggest that diazene JK-279 may, due to reduction of GSH, induce apoptosis in human cervical carcinoma cells.
