Sažetak
Background: Understanding the immunological mechanisms how the host recognizes countless foreign antigens and remains unresponsive to self have opened many questions in the field of immunological tolerance, but the answer is still remain unclear. The most specific marker that distinguishes regulatory from conventional T cells is forkhead box transcription factor (Foxp 3) (1). Predominant cell types that expressed Foxp 3 are CD4+CD25+ and characteristic of subpopulations which are Foxp 3 negative are their regulatory function lacked. Natural Tregs have the possibility to suppress multiple cell types involved in immunity and inflammation by inhibition the proliferation, immunoglobulin production, the blocking of NK and NKT-cell cytotoxicity, as well as, the function and maturation of dendritic cells. The most common use of Tregs is described in the prevention of autoimmunity (2) in possibility to moderate transplantation tolerance and in allergic diseases (3). Urban air pollutants are known to contribute to diminished immunological tolerance, contributing to increasing prevalence of environmental diseases and multiple chemical sensitivity or other related syndromes, as a result of an abnormal immune response based on environmental damage of lymphocyte subsets. The aim of this study: was to analyse the characteristics of human peripheral blood lymphocytes (phenotypic profiles, cytotoxicity against human NK sensitive cell line and the cytolitic ability with measuring the percentage of perforin positive cells), of people from industrial areas and correlate with them from rural ones. Patients and methods: A total of 46 inhabitants were investigated, devised in two groups: from oil industry area and from rural ones. None of inhabitants was taking any systemically administered medications for at least 3 months before testing. Phenotype of lymphocytes and perforin expression were analyzed using intracellular and surface immunofluoresecncy and flow cytometry (FACSCalibur). Statistical analysis was done in computer program Statistica 7.1 (StatSoft, Inc., Tulsa, OK, USA). Results: People from industrial area have significantly increased NK, NKT and Treg cells and diminished values of cytolitic molecule perforin in peripheral blood lymphocytes, which correlate with decreased cytotoxic potentials of lymphocytes. 59, % inhabitants from industrial areas have moderate restriction/obstruction spirometry, compared to 15% inhabitants from rural areas. Conclusion: Environmental factors-induced immunological changes may contribute to develop environmental diseases. Our data try to elucidate the positive effects of environmental factors on the developing of COPB and asthma. Tregs have the ability to suppress allergic immune response induced by urban air pollution and their role in environmental diseases is marked.
