Sažetak
Abstract: When fungi are grown in submerged cultures a large number of factors contribute to development of different morphological forms, ranging from disperse mycelial filaments to densely interwoven mycelial masses, known as pellets. Pellet cultures form aggregates of hyphae as they grow, exhibit low viscosity, approach Newtonian flow behavior and do not adhere to any part of the bioreactor. In this work, a process of laccase production and growth of T. versicolor pellets in submerged culture was performed. Experiments were carried out in a batch mode on rotary shaker in Erlenmeyer flasks and in a bioreactor. The influence of initial concentrations of simple sugars as sources of carbon (glucose and sucrose), yeast extract and peptone as sources of nitrogen, as well as the concentration of other media components (microelements, Tween 80, salts) on sugar consumption, pellet’ s production and laccase activity was investigated. In experiments carried out in a bioreactor the pulp and paper industry waste was used as inductor for laccase synthesis. The influence of mixing, dissolved oxygen concentration and pH on laccase activity was investigated.
