Sažetak
Here we present electron paramagnetic resonance (EPR) using DMPO spin trapping in the study of antioxidant potency of two different ganglioside types. Namely, exogenously administrated gangliosides have been observed to affect many cellular functions. It has been suggested that gangliosides might act as antioxidants in lipid peroxidation [1]. In this context we have used Fenton type of reaction and xanthine/xanthine oxidase to model generation of hydroxyl and superoxide radicals, respectively [2]. The analysis of EPR signal intensity indicated that the content of hydroxyl radical spin adducts was lower in the presence of gangliosides at concentrations above their respective critical micellar concentrations in comparison with the samples without gangliosides. We show that the primary mode of ganglioside actions is not due to direct scavenging of hydroxyl radicals, but rather to the inhibition of their formation. Regarding superoxide anion production no experimental evidence for ganglioside scavenging effect was observed. The effects of gangliosides on the radical yield in the cell free system are discussed in relation with the results obtained with human polymorphonucelar leukocytes (PMNs) stimulated by phorbol myristate acetate (PMA).
