Sažetak
Superoxide dismutase (SOD), a superoxide anion (O_2-) scavenger, is a constituent of human spermatozoa, but its specific role sould be further elucidated. The aim of this study was to determine SOD-like activity ans superoxide anion production in spermatozoa of asthenozoospermic samples characterised by reduced motility (above 20 million spermstozoa/ml ; below 40% motile sperms). Normozoospermic samples with above 40% motile sperm served as controls. Percoll gradient fractionation was used to separate the spermatozoa with high motility and competent function (80% fraction) from those with diminished motility and poor functional capacity(40% fraction). SOD-like activity was measured using the RanSod test and the superoxide anu+ion production was quantified y measuring ferrcytochrome-C reduction. SOD-like activity in 12 asthenozoospermic samples from a 40% Percoll gradient was significantly higher thasn those recorded in the same fraction of 18 normozoospermic samples (3.78 vs 2.19 U/108 spermatozoa ; p 0.05), whereas a non-significant increase was observed in spermatozoa from an 80% fraction (2.68 v 1.46 U/10 8 spermatozoa P 0.5). There was no significant difference in superoxide anion production in both fractions of normo-and asthenozoospermic samples(0.47 v. 0.46 nmol/10 6 spermatozoa in the 40% fraction and 0.75 v. 0.92 nmol/10 6 spermatozoa in the 80% fraction, respectively. Thus, higher SOD-like activity in asthenozoospermia probably indicates increased availability of cytoplasmic enzymes as a consequence of retention of excess cytoplasm during spermiogenessis and is not associated with the level of supweroxide anion. Determination of SOD-like activity in spermatozoa is likely to be a valuable tool for investigating its quality and maturity, especially in preparation for in vitro fetrtilisation.
