Sažetak
Trophoblastic cells from the ectoplacental cone and decidua cells interact in formation of the major part of the rat placenta. The influence of the teratogen 5-azacytidine (5azaC) on the development of the ectoplacental cone and deciduoma during the critical phases of embryogenesis was investigated both in vitro and in vivo. 5azaC (5mg/kg) was administered intaperitoneally to Fischer rat females at the 5th or 8th day of gestation and controls were untreated. On the 9th day of gestation deciduomas were isolated from the uterus. They were dissected under the microscope and the ectoplacental cones (EPCs) extracted. EPCs were cultivated in vitro during three days and their diameters were measured before fixation by the ocular micrometer. After isolation, some EPCs were transplanted under the kidney capsule of the adult rat for 6 weeks. Capacity for proliferation of the decidua cells was detected by immunohistochemistry using two proliferation markers Proliferating Cell Nuclear Antigen (PCNA) and Ki67. PCNA expression was stereologically quantified (numerical density, Nv) and statistically evaluated by Student’ s t-test. 5azaC administered on the 8th day of gestation increased growth of the EPC in vitro. In EPC transplants, ingrowths to the kidney parenchyma were detected consisting of a massive fibrous reactivity and lymphocytic infiltration. It seems that this reaction was more prominent in the case of EPCs treated on the 5th day of gestation. PCNA expression was significantly higher in decidua cells for both 5azaC treated experimental groups and in comparison to controls (p<0.05). Moreover, in the group treated on the 5th day, PCNA expression was significantly higher than in the group treated on the 8th day of gestation. These results show that the periimplantation and perigastrulation periods known to be critical for the embryonic development are also crucial for the development of extraembryonic structures and decidua which are to build the placenta.
