Sažetak
The role of angiogenesis in the pathogenesis of multiple myeloma (MM) is well recognized, however the involvement of myeloma cells and diverse microenvironment elements of bone marrow in this process is not absolutely understand. The aim of this study was to explore the expression of three different cytokines, recognized to be involved in the angiogenesis. The expression was analyzed at the protein level in bone marrows and at the plasma levels of MM patients. Expression of cytokines was correlated with microvessel density (MVD) and clinicopathological characteristics of MM patients. Immunohistochemical analysis was performed for visualization of vascular endothelial growth factor (VEGF), macrophage chemotactic protein-1 (MCP-1), osteopontin (OPN), and endothelial cells (CD34), while ELISA method was used for detection of plasmas levels of VEGF, MCP-1 and OPN. The results demonstrated higher values o MVD in bone marrows with higher percentage of plasma cells infiltrates, in patients with higher clinical stage, no response to therapy and lower overall survival. All three cytokines were detected in various percentage and staining intensity in the myeloma cells, different hematopoetic and stromal cells. Some positive staining was also observed in extracellular matrix of bone marrow. Coexpression of all three cytokines was detected at the protein levels in the myeolma cells and at the plasmas levels of MM patients. When correlated with MVD only VEGF expression demonstrated inverse relationship. Lower expression of VEGF was associated with higher value of MVD. Two others cytokine at the protein and all three at the plasmas levels did not correlate with MVD. Nevertheless plasma VEGF, MCP-1 and OPN concentrations were higher in those patients with higher percentage of tumor cells infiltrates in bone marrows. Present results confirmed the prognostic significance of MVD in MM and also the association of cytokines levels with tumor mass what should be further explored.
