Sažetak
Mre11 and MRX complex play an importan role in the initial phases of repair of double strand breaks (DSBs) in meiosis in all organisms tested today. Mre11 p is assumed to be involved in DSBs resection and removing of the Spo11 p from that place. In contrast to yeast, worms and some other models where the C-terminus of the protein Mre11 is essentail for the activity of Spo11, we propose that C-terminus has no such activity in plants. To prove this hypothesis we obtained three insertional mutants of AtMRE11 gene with insertions in the 9th intron (mre11-3), 18th intron (mre11-4) and 19th exon (mre11-2). In contrast to mre11-3 and mre11-4, which exhibited severe growth defects and were completely sterile, mre11-2 mutant had normal phenotype and was fully fertile. A detailed cytogenetic analyses of meiosis in mre11-3 and mre11-4 mutants showed abnormailities in meiosis: absence of chromosome pairing in the initiala stages of meiosis and massive genome fragmentation, after the leptotene stage. In spite of that, meiosis in both mutants progressed till the end and resulted in formation of polyads containing microspores with unqueal amount of DNA. Meiosis in mre11-2 was normal. This finding suggets that C-terminus of the Mre11 protein in not required for the function of Mre11 and MRX complex in Arabidopsis. It also indicates that meiotic functions of Mre11 protein may not be as conserved among eukaryotes as has originally been asumed.
