Gene therapy is a very promising approach of curing diseases on molecular level. DNA vaccines comprise one category of gene therapy and it has been shown that plasmid DNA encoding antigens induce both cell-mediated and humoral, Th-1-biased immune responses. Their efficacy greatly depends on efficiency of DNA transfer into cell and subsequently nucleus. A group of mostly used non-viral vectors facilitating this process are cationic liposomes. Cationic liposomes complex DNA and deliver it into the cell. Physical and chemical properties of liposome vectors have been shown to influence their transfection efficiency in vitro and in vivo. The aim of this work was to establish DNA mumps vaccine using plasmid encoding haemagglutinin and cationic liposome vector. In order to optimize properties of complexes for delivery of DNA, complex size, charge and lipid composition have been systematically investigated in vitro using plasmid encoding green fluorescent protein which enabled us to easily quantify transfection efficiency by flow citometry. Subsequently, transfection efficiency results have been confirmed with plasmid encoding haemagglutinin by detection with anti-haemaglutinin antibodies and also flow citometry. Our results reveal that the transfection efficiency in vitro is enhanced by sizing down liposomes and by using very high positive overall charges. In addition, transfection efficiency has been shown to substantially vary with lipid composition of vector and with the cell type used.
