Sažetak
Objectives: Objective of this study was to determine the degree of DNA molecule damage in different time intervals after ozone exposure. Methods: In this study alkaline comet assay cytogenetic technique modified by applying formamidopyrimidine-DNA glycosylase (Fpg) was used to enable detection of DNA damage caused by oxidative stress. To evaluate genome damage primary human lymphocyte cultures were used. Samples were then analyzed using Comet assay II automatic digital analysis system (Perceptive Instruments Ltd, Suffolk, Halstead, UK) to measure the tail moment and tail intensity (percentage of DNA that migrated into the tail). Results: In the Table 1, tail intensity and tail moment represent a degree of damage after the cells being treated with buffer without the endonuclease (labeled buffer) and the second column (labeled Fpg) represent a degree after treatment with Fpg. So, if there is an oxidative damage, the difference between parameters measured with Fpg and only buffer represent exact intensity of oxidative damage. Conclusion: Even though there is some initial DNA damage, two hours following ozone application there are no differences observed between treated and control lymphocytes, indicating that the induced lesions have been efficiently repaired.
