Sažetak
Peptide-D is a pentapeptide recently defined by means of SCA method. This peptide is involved in Ca2+ signalling in mammalian cells. We investigated Peptide-D and its complementary peptide (Met-enkephalin/Peptid-M) that belongs to the family of neuropeptides. The effects of the peptides on peripheral blood lymphocytes were analysed in vitro with respect to their influence on cell cycle, proliferation, apoptosis and mutagenesis. In vivo toxicity and cytoprotection were evaluated on the rat and mice model. Peptide-D and its complementary motif were analysed by one- and two-dimensional 1H and 13C NMR spectroscopy. Their fragmentation was followed by laser desorption mass spectroscopy. Spectroscopic findings and in vitro and in vivo studies contributed to the understanding of the physiologic and pathologic functions of the calpain-calpastatin system and related Met-enkephalin-FKB3 interactions. The results are discussed in the context of several ocular, nervous system and rheumatic diseases including: cataract, uveitis, optic neuritis, multiple sclerosis, Alzheimer's disease and rheumatoid arthritis.
