Sažetak
"Split" vaccine against infectious bonchitis, prepared in Veterina Ltd., contained the IB virus propagated in embrionated hen eggs from specified pathogen-free (SPF) flocks. The virus is inactivated with 0.04% formaldehyde, and entrapped in the vaccine that was prepared in form of water-in-oil-in-water emulsion (WOWE). WOWE were tested for physical stability (as regards droplet size and distribution) after 3 and 6 month of storage at room temperature what is the stress condition for vaccines. The technology of image analysis of microscopical picture by application of the two systems for particle characterization was employed ; a digital microscopy camera system Axioscop with computer software for visualization and interactive morphometry, and Olympus BH-2 with Optomax V equipment for automatic measurements. After 6 months storage at stress condition (20°C), the mean diameter of WOWE droplets changed from 1.0653 to 1.2143 micrometers, but the distribution remained statistically similar. During the observed period the values of mean droplet diameter slightly increased from 100 to 200 nm. The results obtained by image analysis microscopy techniques demonstrated that tested WOWE remained very stable after 6 months storage under unsuitable conditions.
