Sažetak
The Hh-Gli signaling pathway also known as 'Hh-Ptc' or 'Hh/Ptc/Smo' plays a major role in embryonic development and its deregulation is related to developmental malformations and tumorigenesis. Dermoids are tumors that develop from embryonic stem cells and they show elements of both developmental malformations and tumors. Our results contribute to existance of two hypothetical models of abnormal Hh-Gli pathway activation. One model suggests constitutive activation of the pathway within the cell, and doesn not require outside stimulation. The other model suggests abnormal Hh stimulation from the surrounding cells, which causes a strong intracellular response of the pathway. Hh-Gli signaling pathwa related genes (Shh, Ptch, Smo, Gli1, 2 and 3) showed different levels of expression by RT-PCR in different cell lines derived from each primary culture of ovarian dermoids proving their heterogenous nature. In some cases, the high levels of expression of tested genes were efficiently bolcked with cyclopamine, and then the pathway was reactivated with Shh. Additionally, clones that have not responded to cyclopamine, or to reactivation of the pathway support another hypothetical model. Our results contribute to understanding of Hh-Gli signaling pathway activity in carcinogenesis and to development of methods for signal inhibition which will lead to normalizing the function of Hh-Gli pathway, and to tumor inhibition.
