Sažetak
Tolerance mechanisms are responsible for the fetus survival and regulatory T cells (Tregs) seem to have very important role in the tolerance towards the fetus bearing alloantigens. These mechanisms involve the induction of cell death or the development of a no responsive state (anergy) of T cells. Tregs have active suppression mode and may induce peripheral tolerance. Mammalian pregnancy is a state of immunological tolerance, which’s lost, may cause immunological pregnancy complications, such as spontaneous abortion and pre-eclampsia. NKT cells have characteristics of NK cells and classical T cells, which are involved in control of antitumor, autoimmune, allergic and antimicrobial responses. They regulate immune response by fast excretion of large amount of Th1 and Th2 cytokines. Activated NKT cells have a possibility to modulate quantitatively and qualitatively Treg function through IL-2-dependent mechanisms. Furthermore, Tregs can suppress the proliferation, cytokine release and cytotoxic activity of NKT cells by cell-contact-dependent mechanisms. Toll-like receptors (TLRs) have a pivotal role in innate immune response and are expressed by trophoblast in normal full-term placenta. To examine the effects of in vivo administration of natural TLR2 ligand-peptidoglycan monomer linked with Zn (PgMZn) during murine pregnancy on phenotypic profile and cytotoxic characteristics of maternal liver, spleen, thymus and uterus during syngeneic pregnancy on C57bl/6 mice. Pregnancy increased the values of Tregs and NKT cells in maternal liver, augmented cytotoxicity against YAC-1 tumor line (NK-sensitive target), syngeneic thymocytes and P815 (LAK-target). PgMZn administration potentates these effects. The data point the interplay between NKT and Tregs signify that liver and bacterial products PgMZn are important players in moderating of innate immune response during pregnancy.
