Sažetak
Peroxidases are enzymes that catalyze oxidoreduction of cell substrates by hydrogen or organic peroxides. They are hem, flavine and selenoproteins, according to associated prosthetic group. Peroxidases have important function in biochemical and physiological processes in plants and animal organisms they are included in protection from oxidative stress and infections and in the process of hormone synthesis. Glutathione peroxidases are selenoproteins and their role in primary antioxidans defense is proved. Using reduced glutathione glutathione peroxidases remove free radicals and peroxides. Laboratory research results showed that peroxidases can use guaiacol as substrate in vitro. Coumarins are important group of compounds with physiological role not cleared completely. Natural coumarin sources are green plants and they are present as isocoumarins, pyranocoumarins and furanocoumarins. Coumarins have influence on physiological and biochemical processes in plants, although some coumarin types showed anticoagulation property and inhibition on huge number of human cancer cells. Ethyl 2-(3, 6-dibromo-7-hydroksi-2-oxo-4-chromen-4-yl) acetate (K2) is 7-hydoxycoumarin, is water insoluble, but soluble in ethanol and dimethylsulphoxide (DMSO). Purpose of this experiment was to find out how K2 affect glutathione peroxidase activity in enzyme assay, where guaiacol was used as a substrate. Peroxidase activity is expressed as A min-1. Results of our experiments show that K2 has an inhibitory effect on peroxidase activity, which increase with higher K2 concentration. Researches of scopoletin and his affect on horseradish peroxidases show that scopoletin acts as a competitive inhibitor whose effect could be neutralized if substrate concentration is rised. Future research could be directed on type of inhibition glutathione peroxidase, (reversibile or irreversibile).
