Sažetak
Problem: The study was aimed to analyse phenotype and functional properties of the first trimester normal human pregnancy decidual CD1a+ dendritic cells (DC). Method of study: Decidual mononuclear cells (DMC) are obtained by enzimatic digestion and gradient density centrifugation. The phenotype of CD1a+ DC (differentiation and maturation markers, as well as NK cytotoxicity receptors) was analysed immediately after the isolation and in both: adherent and non-adherent DMC fraction obtained after the 18-hour culture in vitro. In freshly isolated DMC, cell permeabilisation method with simultaneous surface CD1a labeling was used for cytokine and cytolytic mediator detection. Results: Decidual CD1a+ cells are lin-1 negative and show mostly myeloid, immature DC phenotype according relatively low CD83, CD80, CD86, HLA-DR and high CD14 molecule expression. There is no significant difference in the phenotype of CD1a+ DC between adherent and nonadherent fraction. CD1a+ cells from freshly isolated suspensions express mostly IL-15 and IL-18 cytokines, the percentage of IFN- + and TNF- + cells was approximately 2 times lower and the percentage of IL-10 producing cells was negligable. CD1a+ cells are equiped with short and long term cytolytic mediators, as well as surface NK cytotoxicity receptors. Conclusion: Mostly immature CD1a+ DC represent a source of pro-inflammatory cytokines and could be involved in cytotoxicity reactions in decidua according to their equipment with cytolytic armentarium.
