Sažetak
We have developed novel oil-based adjuvant formulations by incorporation of immunostimulating peptidoglycan monomer (PGM) into Montanide ISA720 and ISA206 oil-based adjuvants. So far they have shown excellent adjuvanticity in improving humoral immune response in rabbits to several tested antigens. The aim of this study was to investigate the influence of novel formulations on Th1/Th2-type of immune response with respect to the induction of IgG isotypes, IgG1 and IgG2a. CBA (H-2k) and NIH/OlaHsd (H-2q) mice were immunised s.c. with ovalbumin (OVA) as an antigen adjuvanted in ISA720 and ISA720+PGM or ISA206 and ISA206+PGM. Positive control group was immunised with OVA+CFA/IFA. IgG as well as IgG1 and IgG2a immune responses were determined by ELISA after the first and the second booster. For Th1/Th2 evaluation the ratio of IgG1 to IgG2a (IgG1/IgG2a) as specific markers of Th2 and Th1-type of immune response, respectively, was calculated for each serum in experimental groups and compared to the ratio of no adjuvant-treated group. Two adjuvants tested exhibited different effects in directing specific immune response depending on mice strain used in the study. The IgG1/IgG2a of no adjuvant-treated group in both mice indicated Th2-biased immune response specific for OVA. In CBA mice both ISA adjuvants induced the switch towards Th1, with ISA720 being stronger one and also very similar to CFA. In NIH/OlaHsd mice ISA720 stimulated Th1-biased immune response also, while ISA206 induced the switch towards Th2. The influence of PGM incorporation didn't seem to influence the direction of the immune response already established by oil-based adjuvants.
