Sažetak
In our previous studies we have demonstrated the interactions of ovalbumin (OVA) with lipids in liposomal membranes [1] and the effects of OVA encapsulated in liposomes on the immune response in mice [2]. The aim of this work was to investigate the size and zeta potential of large multilamellar liposomes with incorporated ovalbumin using dynamic light scattering and fluorescence microscopy. Multilamellar cationic liposomes consisting of phosphatidylcholine (PC): dioleoylphosphatidylethanolamine (DOPE) : 1, 2-Dioleoyl-3-trimethylammoniumpropane (DOTAP) and negatively charged liposomes of phosphatidylcholine: cholesterol (Chol): dicetylphosphate (DcPh) were prepared by the thin film hydration method. Different OVA quantities and different buffers for encapsulation OVA in liposomes were used. The rodamine B and fluorescein-isothiocianate were used for fluorescence measurements. The results revealed that the size of cationic liposomes in PBS increases linearly with increasing amount of OVA. The zeta-potential of examined liposomes preparations was slightly different.
