Sažetak
Under the ongoing project, Reduction of environmental risks, posed by emerging contaminants, through advanced treatment of municipal and industrial wastes (EMCO ; 6thFWP INCO CT 2004-509188) we applied four bioassays to analyze biological relevant changes in contaminants concentration and effect of waste water samples before and after treatment. Specificity of the project is that it searches for so-called “ emerging” or “ new” contaminants that are unregulated and with a lack of knowledge of their pollution potential. During the two campaigns, 1st campaign (October, November 2004) and 2nd campaign (April-May 2005) waste water samples were collected at the locations indicated in Table 1 and 2. Immediately after collection samples were extracted and methanol elutes of waste water were dried under the stream of nitrogen gas. Final residuals were dissolved in DMSO to achieve 2.000x concentration (samples from the 1st campaign) or 3.000x concentration (samples from the 2nd campaign) from the original water sample. Before testing all samples were serially diluted in solvent and/or in corresponding media. Estrogenicity of the samples was measured by using yeast estrogen screen (YES) assay to detect ER agonists. MXR- inhibiting properties of samples was determined by using a P-glycoprotein (Pgp) inhibition assay. CYP1A induction potential of environmental samples was determined by measuring EROD activity in PLHC-1 fish hepatoma cell line. Overall toxicity of the samples was measured by determination of growth inhibition rate of unicellular algae Selenastrum capricornatum.
