Sažetak
"Split" vaccine against infectious bronchitis, prepared in Veterina Ltd., contains the IB virus propagated in embrionated hen eggs from specified pathogens-free (SPF) flocks. The vaccine was prepared in form of water-in-oil-in water emulsion (WOWE), with alow content of mineral oil. WOWE were tested for physical stability after 3 and 6 month storage at room temperature what is the stress condition for vaccines that should be kept at temperature of 2 - 8oC. The technology of image analysis of microscopical picture by application of two systems for particle characterization was emploed. A digital microscopy camera system Axioscop with computer software for visualisation and interactive morphometry, and Olympus BH-2 with Optomax V equipment for automatic measurments, were used. In all tests approximately 10000 droplets of each sample were examined. The results demonstrated that tested WOWE (the mean diameter of droplets changed from 1.065 to 1.214 micrometers /after 6-moths storage at stress condition/, and from 100 to 200 nm /during the observed period/)remained very stable after 6-motnhs storage under unsuitable conditions.
