Aim. To investigate ganglioside expression in lymphoid (spleen, thymus, and lymph nodes) and other tissues (lungs, brain, muscle, liver) of mice deficient in beta2-microglobulin (B2MG), component of the histocompatibility class I antigens (MHC I). Methods. Gangliosides were isolated from C57BL/6 mice homozygous (-/-) or heterozygous (-/+) for the B2MG gene knockout and separated by high pereformance thin-layer chromatography (HPTLC). Individual gangliosides were visualized using resorcinol stain and HPTLC immunostainig overlay technique with polyclonal antibodies specific for GM3(Neu5Ac), GM3(Neu5Gc), GalNAcGM1b, and nLc4. GM1b-type gangliosides were detected after V. cholerae neuraminidase treatment with a specific anti-Gg4 antibody. Neuraminidase pretreatment was also used to detect gangliosides IV3nLc4 and VI3nLc6. GM1a and Gal-GalNac-GM1b gangliosides were detected with choleragenoid. Individual HPTLC bands were scanned with a CD60 scanner to quantify the differences between -/- and -/+ mice. Results. GM3(NeuAc) was four times and GM3(Neu5Gc) three times more abundant in the lungs of -/+ compared to -/- mice. Two bands of GM1b, GalNac-GM1b, and Gal- GalNac-GM1b, respectively, were expresses in the lungs of -/+ mice, but not in the lungs of -/- mice. Gangliosides of the neolacto-series were several times more abundant in the muscle, spleen, thymus, and lymph nodes of -/+ mice than of -/- mice. Conclusion. Our study provides in vivo evidence that B2MG is involved in the acyusition of distinct ganglioside profiles of different mouse organs.
