Various tests in aquatic organisms have been developed for the detection and identification of toxic substances. In the present study, we utilized the alkaline comet assay to conduct an environmental assessment of water quality using a benthic fish species. The objective of the study was also to evaluate the potential usefulness of Cobitis elongata as a biomarker species. The alkaline single cell gel electrophoresis, also known as the comet assay, is a rapid, simple and sensitive technique for measuring DNA strand breaks in individual cells. Sampling was conducted at two sites: Ivanja rijeka municipal waters of the City of Zagreb and the Kupa River clean waters in the vicinity of the City of Karlovac. The alkaline comet assay was performed on nucleated erythrocytes. The amount of DNA damage in cells was estimated from three different parameters: comet tail length as the extent of genetic material migration, tail intensity (% of DNA in comet tail) and tail moment. The results demonstrated that fish captured from Ivanja Rijeka displayed relatively higher levels of DNA damage compared to those captured from the Kupa River. The results suggest a genotoxicity of the aquatic environment at Ivanja Rijeka. This study confirmed that the comet assay applied on the fish erythrocyte is a useful tool in determining potential genotoxicity of water pollutants. The results obtained also suggest that Cobitis elongata provides adequate sensitivity to be utilized as a potential biomarker species in the monitoring of water pollution and environmental risk assessment.
