Sažetak
Problem: The aim of our study was to investigate the change in conformation of Ld molecules during the endocytic trafficking. Material and methods: The expression of Ld molecules was determined by flow cytometry, confocal microscopy and immunoprecipitation. Misfolded conformation was achieved by short acidification. Localization of molecules in membrane domains was determined by Triton X-100 treatment. Results: Non-conformed Ld molecules, in contrast to conformed, localize in the lipid rafts. Although the majority of conformed Ld molecules were disintegrated shortly after endocytosis, non-conformed Ld are directed to an endosomal retention compartment. However, after inhibition of endosomal acidification, they colocalize to the large extent. Treatment of cells with filipin retards recovery of conformed molecules at the cell surface, and in cycloheximide treated cells increases the expression of non-conformed Ld molecules. Conclusion: Ld molecules change their conformation in intracellular vesicles after endocytosis and probably provide a basis for alternative presentation.
