Sažetak
Organic anion transporter Oat5 (Slc22a19) is a member of multispecific drug transporter family that resides in the kidney (K) epithelial cell plasma membrane and contributes to the excretion and/or reabsorption of various endogenous and exogenous organic anions. Oat5 has been cloned, defined as an organic anion/dicarboxylate exchanger, identified in rat and mouse K with similar levels of mRNA expression in females (F) and males (M), and localized to the brush-border (BB) of proximal tubules (PT) in the outer medulla. We have re-investigated the expression of rat Oat5 (rOat5) mRNA by RT-PCR in the K tissue zones and localization of its protein by Western blotting (WB) of isolated cell membranes and by immunocytochemistry (IC) in tissue cryosections from adult intact and gonadectomized M and F, castrated M treated with various sex hormones, and prepubertal intact M and F rats. In adult rats, the expression of rOat5 mRNA was predominant in the outer stripe (OS), where it exhibited: a) gender differences (F>M), b) upregulation in M and downregulation in F by gonadectomy, and c) strong downregulation in testosterone- and slight upregulation in estradiol- and progesterone-treated castrated M. In WB of isolated K membranes from variously-treated rats, a polyclonal anti-rOat5 antibody labeled in non-reducing conditions a single protein band at ~70 kDa which was stronger in F and showed the density pattern comparable to that of mRNA expression. By IC, the BB of cortical convoluted PT in intact M and F was weakly stained, whereas the BB of PT S3 segments in the OS and medullary rays were stained brightly and much stronger in F. In variously-treated M rats, the pattern of BB staining intensity in S3 matched the mRNA and WB data. In prepubertal rats, the expression of rOat5 protein in PT was strong and similar in M and F. In conclusion, the rat renal Oat5 is largely localized to the BB of PT S3 segments where it exhibits gender differences (F>M) that appear after puberty due to strong androgen inhibition and weak estrogen and progesterone stimulation.
