Sažetak
Intracellular assessment of cytokine at th single cell level by flow cytometry has recently become available. Several critical parameters such as the cell type used, mode of activation, and intensity and timing of cytokine generation should be adjusted for each cytokine tested. We have established protocols for the evaluation of cytokine production in T cells and monocytes after short term culture with polyclonal activators in the presence of protein secretion inhibitors. The potential use of this method in the clinical evaluation of immune status in patients suffering from allergic disease and respiratory syncycial virus infection demonstrates that intracellular cytokine flow cytometry can be used as a routine test in clinical immunology laboratory.
