Sažetak
In the pathogenesis of secondary type of immuno suppression that often may be seen in patients operated in general endotracheal anaesthesia the participation of various mechanisms has been proposed to exist.They include toxic action of anaesthetic itself, disturbance of neuroendocrinoimmune homeostasis, provoked by anaesthesia or by operative stress and local-trauma induced events which lead to the release of the products of arachidonic acid metabolism and different cytokines from damaged tissue. In an attempt to investigate this type of immunosuppression we tried: 1)to establish an animal model which mimics the clinical situation and 2) to test the therapeutic possibilities of some agents known to affect various points in the regulation of immune response. For this purpose we estimated humoral(by standard plaque forming assay) and cellular immunity (by determination of rejection time of an allogeneic tumor) of mice which were subjected to isolated or combined effects of halothane anaesthesia and operative stress.Immunosuppressed mice were than treated by:a)insulin like growth factor (IGF-I) ; b)indomethacin and c)peptidoglycan monomers, extracted from walls of Gram (+) bacteria.The data have shown that immunosuppression may be induced by halothane anaesthesia alone and partially corrected by tested substances.The analysis of cellularity and phenotypization of cells during these treatments point to immunorestoration and/or contrasuppression as a mechanisms of action of tested agents.
