Sažetak
Abstract Introduction: In a recent literature ultrasonic nebulization system (UNS) had been proposed for delivery of poorly water-soluble drugs into the lungs. This study aims to analyse the use of UNS system for delivery of beclomethasone dipropionate (beclo) in prevention of airway eosinophilia in mice asthma model. Materials and methods: A Sono-Tek ultrasonic spray system was used for production of aerosol from an ethanol solution of beclo. The aerosol stream is then passed through a drying column (TSI) containing an annular ring of charcoal and deionizator (TSI). Described system successfully produced dry beclo particles of inhalable size (approximately 1.5  m). Particles were delivered to BALB/c mice placed in whole-body exposition chamber one hour before single intranasal challenge of ovalbumin (OVA) to BALB/c mice that were previously sensitised to OVA (n ≥ 8). Efficacy of beclo delivery was evaluated by determining relative number of eosinophils in BALF 48 hours after challenge. Results: Size of dry powder particles produced by USN system increased as the concentration of beclo in aeroslized solution increased. After determining particle size, 1 mg/ml solution of beclo was chosen as an optimal concentration for inhalable dry powder particle production. Beclo applied through UNS system in the calculated dose of 100  g/kg (b.w.) significantly decreased relative eosinophil number in BALF for almost 90% (from 42.86% to 3.89% eosinophils in BALF ; t-Test p < 0.05). Conclusions: Beclo applicated with UNS system significantly reduced eosinophilia in BALB/c mice asthma model. Thus, the results of present study show that UNS system can be used for rapid screening of respiratory delivery of drugs and test compounds.
