Sažetak
Lysosomes concentrate a wide range of contaminants from within the environment, including lipophilic xenobiotics and metals, that may cause changes in the integrity of their membranes (1, 2). Retention of the cationic probe neutral red within the lysosomal compartment was used as a measure of damage to the lysosomal membrane. The acid environment of lysosomes is maintained by the membrane Mg2+-ATPase dependent H+ ion proton pump (3). The neutral red retention assay (NRR) reflects the efflux of the lysosomal contents into the cytosol following damage to the membrane and, possibly, impairment of the H+ ion pump (4). Reductions in lysosomal membrane stability in digestive cells have already been recorded after experimental exposure of marine molluscs to copper (5, 6, 7) and cadmium (8, 9). The objective of this research was to investigate possible use of haemocyte lysosomes of freshwater bivalve Dreissena polymorpha as cellular marker of environmental pollution. The neutral red retention assay was adapted to assess the effects of cadmium and copper on haemocytes of D. polymorpha.
