Sažetak
The reflux laryngitis is the main extraesophageal manifestation of gastroesophageal reflux disease (GERD). Gastroesophageal reflux disease is defined as chronic disease with mucosal damage produced by the abnormal reflux of gastric contents into the esophagus. Extraesophageal GERD complications, such as reflux laryngitis, develop when some of the acidic refluxate in the proximal esophagus enters the lower or upper airways because of comprised defense mechanisms and it results from abnormal exposure of laryngeal mucosa to acid refluxate. Experimental studies have shown that pentadecapeptide BPC-157 have protective effect on esophageal mucosa in an animal model of GERD. The aim of the study was to evaluate the effect of gastric pentadecapeptide BPC-157 therapy on rats with reflux laryngitis. The study was done on fifty adult female Wistar rats and they were randomly assigned into five groups, four experimental groups (treated with 10 µ ; g/kg and 10 ng/kg pentadecapeptide BPC-157, intraperitoneally and intragastrically) and control (treated with 0.9% saline). All animals underwent an operation to implant a polyethylen tube, through the stomach incision, connecting stomach and nasopharynx through the esophagus, causing the gastroesophageal and laryngopharyngeal reflux. All rats were divided into two groups, sacrificed at two and four weeks after the operation and examined for macroscopic and pathohistologic changes of the laryngeal mucosa. The macroscopic grades of inflammatory changes of the laryngeal mucosa of the treated animals, for both concentrations (10 µ ; g/kg and 10 ng/kg), were significantly better than that in the control (p<.01) and the microscopic grades of inflammatory cells accumulation of the laryngeal mucosa in experimental group was significantly better than that in the control group (p<.05). There was found significant correlation between the grades of pathohistologic mucosal changes and macroscopic changes of laryngeal mucosa (p<.05). The results suggest that the gastric pentadecapeptide BPC-157 may have protective effect on reflux laryngitis in gastroesophageal reflux disease.
