Sažetak
Tissue Typing Centre Zagreb is involved in BMT program from its very beginning. In the period 1983-2004, a total of 1537 patients underwent the program of searching of related bone marrow donors. For that purpose all patients and 6122 of their family members were tested for HLA polymorphism. All individuals were typed for HLA class I and class II alleles by serology until 1992, when DNA typing techniques (PCR-SSOP and PCR-SSP) were introduced. The present algorithm is: i) HLA class I typing by serology and additionally by DNA using low resolution HLA class I typing in the case of ambiguous results or homozigosity ; ii) HLA class II typing by DNA routinely using low resolution HLA class II typing and high resolution HLA class II typing in the case of homozigosity ; iii) MLC test. In total, 554 (36.1%) patients had HLA genotypically identical sibling donor, while 14 (0.9%) patients had HLA fenotypically identical parents. Eight patients (0.05%) received bone marrow from family donor with one mismatch on HLA-A or HLA-B locus Nineteen patients underwent unrelated BMT preceded with the following HLA typing protocol: i) recipient: class I (A, B) typing by serology and by low/high resolution DNA typing when necessary and class II (DRB1, DQB1) typing by high resolution DNA typing ; ii) confirmatory HLA typing of donor iii) MLC test.
