Sažetak
The T-2 toxin belongs to an important class of naturally occurring toxic compounds produced by fungi of the genera Fusarium spp. Trichothecene mycotoxins produce a variety of toxic symptoms in humans and animals, including mortality, skin and gastrointestinal irritation or necrosis, diarrhoea, vomiting and feed refusal, decreased body weight gain, damage to haematopoietic systems in bone marrow, spleen, thymus and lymph nodes and immunological alterations. As potent inhibitors of protein, DNA and RNA synthesis they are especially toxic to tissues with a high cell division rate. Leucocytes and the immune system are considered as the primary target for trichothecenes. The Comet assay is a sensitive and rapid microgel electrophoretic technique that has been used for assessing levels of DNA damage in individual cells. The purpose of this study was to estimate the application of alkaline comet assay for detection of primary DNA damage in chicken blood cells exposed to T-2 toxin at different time intervals. Parameters of comet assay (tail length and tail moment) have shown increase un level of damage of cells in time dependence compared to the control. According to our results the alkaline comet assay can be used for estimation of DNA damage of chicken leucocytes exposed to T-2 toxin.
