Sažetak
The alkaline comet assay was used to assess primary DNA damage in peripheral blood leukocytes of ten patients who received radiotherapy after were surgically removed different solid tumors. This study included patients with head and neck cancer, prostate cancer, endometrial cancer, lung cancer, breast cancer, and brain and testicular tumour. The levels of DNA damage were evaluated in four blood samples taken before and after receiving the first dose of radiotherapy, in the middle of radiotherapy, and after the last dose of radiotherapy. Two main comet parameters, namely the tail lenght and tail moment were measured. The results indicate inter-individual differences in DNA damage in peripherral blood leukocytes between cancer patients before radiotherapy. After the first dose significantly increased levels of DNA damage ewere record in all cancer patients compared to theit baseline levels. Specific patterns of DNA damage were recorded in samples analysed in the middle of radiotherpy and after receiving the last dose, indicating the possibility of adaptive response in some patients. Our results indicate that radiotherapy is accompanied by significant DNA damage in peripheral blood leukocytes. They also confirm damage in peripheral blood leukocytes of cancer patients.
