Sažetak
The aim of our study was to evaluated the influence of exposure to low-level 935 MHz microwave irradiation on proliferation and division kinetics, colony forming ability, viability Chinese hamster lung cells, cell line V79.Previously prepared samples of cells in culture were exposed for 1, 2 and 3 hours to the 935 MHz continues waves frequency field in Gigahertz Transversal Electromagnetic Mode cell (GTEM-cell). As a signal generator was used Hewlett Packard HP 5675A. The electric field strength was 8, 2 +/- 0, 3 V/cm, with the specific absorption rate (SAR) average 0, 12 W/kg. To determine cell growth, V79 samples were plated in concentration of 1x104 cells per milliliter nutrient medium. Cells raised in a humified atmosphere at 37 °C in 5% CO2. Cell proliferation kinetics was determined by cell counts for each hour of exposure during five post-exposure days. To identify colony-forming ability, cells were cultivated in concentration of 40 cells/ ml medium and incubated as above. Colony forming ability for each hour of exposure was defined by colony counts on the 7th post-exposure day. Trypan blue exclusion test was used to determine cell viability. The determine extent of cell division kinetics alteration, cells were plated in 25 cm2 flasks, exposed for 1, 2 and 3 hour. After 1 day cultivation slides were prepared and stained by Giemsa. Mitotic index was determinate by ratio of dividing cells/total cells. Growth curves of sham and RF-exposed cultures showed small but significant differences between the groups. Exposed cell samples showed significant decrease (p<0.05) after 3 hours of exposure on the 3rd post-exposure day. The cell doubling time was 16.2 hours and did not differ between the groups. The ability of V79 cells to form colonies was not affected by exposure. The data also indicated no significant differences between RF- and sham-exposed cells with respect to mitotic indices and cell viability.It is concluded that 935 MHz low-level radiation electric field might affect cell proliferation kinetics without influence on cell division kinetics.
