Sažetak
The aim of this study was to find out whether previously observed inconsistencies regarding the sensitivity of males and females to GABA-A receptor antagonists could be explained by different routes of drug administration. Male and female Wistar rats and male and female C3H mice were used. Antagonists of GABA-A receptors were given by constant i.v. infusion. Mice failed to show sex differences in doses of picrotoxin producing the first myoclonic twitch. Male mice were more sensitive to bicuculline and male rats to picrotoxin and bicuculline administration. This, together with our previous data showing that following i.p. administration female rats are more sensitive to picrotoxin and male mice to picrotoxin and bicuculline, indicates that both mice and rats show sexually different pharmacodynamic responses to bicuculline. Furthermore, mice show sex differences in pharmacokinetics and rats in pharmacokinetics and pharmacodynamics of picrotoxin.
