Sažetak
In plasma, a part of circulating growth hormone (GH) is bound to the growth hormone binding protein (GHBP). In order to study the role of free GH and GHBP in cases when GH concentrations are drastically elevated (acromegalic patients), we measured GH concentrations and GHBP binding capacity and affinity before, during and after hypophysoectomy of acromegalic patients. The values were obtained from 7 acromegalic patients who were operated by transnasal hypophysoectomy in our hospital. Plasma samples were kept at -20 oC for a period of 1-2 years. GH concentrations were measured by radioimmunoassay, while GHBP binding parameters were obtained by gel-filtration of plasma samples which were preincubated with 125I.GH. GH values of plasma samples which were elevated in patients with acromegaly (>15 ug/L), dropped significantly (<2 ug/L) 24 hours after the operation. GHBP binding parameters, after correcing GH values, were unchanged in patients with acromegaly. Additionally, from GH concentration determinations in plasma samples separated by gel-filtration, we showed that during and after the operation free GH disintegrated much faster in plasma than bound to GHBP. Our results indicate that (a) GHBP binding parameters (affinity and capacity) are unchanged in the plasma when pathological concentrations are present for a prolonged time and (b) the main role of GHBP is probably to protect GH from plasma proteases.
